The cultured JTC-3 cells were floated in phosphate buffered solution, kept in polystyrene tube that was rotated for 2.5 rmp in 37°C degassed water, and exposed to the continuous wave ultrasound with the frequencies of 1, 2 and 4 MHz. The ultrasound intensity was SATA 0.1, 0.2, 0.5, 0.8, 1.0 and 2.0 W/cm² in 1 MHz. 3.0 W/cm² intensity was added in 2 MHz. In 4 MHz, the intensity was 1.0 W/cm² or less. The exposure was 5, 10 or 30 min each in every frequencies and intensities. The cell number was counted and the cell colony was formed in flat cell containers after the exposures. Mild increases and decreases of the cell colonies were observed in some instances in the exposure groups comparing to the control. However, no dose-response with the intensity increase or the elongated exposure was observed, whereas irregular and random changes were present. There was no clear suppression of cell colony formation as observed in the effects of anti-tumor substance. From the facts, no effect on cell colony formation was suggested in the exposure to the continuous wave ultrasound in this report.
